Figlia di due fisici di fama mondiale, Meg ha problemi di autostima come tutte le ragazzine della sua età, fatica a integrarsi e cerca disperatamente di farsi degli amici. Non sa ancora di aver ereditato la mente brillante del suo papà e di essere incredibilmente dotata come il fratellino Charles Wallace (Deric McCabe). A peggiorare la situazione interviene la sconcertante scomparsa del signor Murry (Chris Pine), evento che tormenta Meg e che lascia sua madre (Gugu Mbatha-Raw) con il cuore a pezzi. Tuttavia, poco tempo dopo la scomparsa dello scienziato, tre donne dall'aspetto eccentrico e un nome altrettanto peculiare, le signore Whatsit, Who e Which (impersonate rispettivamente da Reese Witherspoon, Mindy Kaling e Oprah Winfrey), spediscono Meg, suo fratello minore e l'amico Calvin (Levi Miller) nello spazio, per salvare il genitore rimasto intrappolato in una faglia temporale.
