Kill Bill Volume 1 racconta le vicende di una donna conosciuta col nome di "The Bride", la sposa (Uma Thurman), che subisce una terribile imboscata durante le sue nozze. Tutti gli invitati, suo marito nonché il figlio che ha in grembo vengono brutalmente sterminati. Dopo essersi svegliata da un coma di 4 anni, la donna intende vendicarsi nei confronti dei responsabili del massacro, che sono per altro vecchi compagni di una vita che si è lasciata alle spalle: si tratta infatti dei membri di un gruppo elitario di killer, guidato dal personaggio di spicco di nome Bill (David Carradine), noti come la Deadly Viper Assassination Squad, o DiVAS. Ognuno di questi efferati assassini ha un nome in codice secondo le varie specie di serpenti velenosi: O'Ren-Ishii (Lucy Liu) è Cottonmouth, Elle Driver (Daryl Hannah) è California Mountain Snake, Vernita Green (Vivica A. Fox) è Copperhead, e Budd (Michael Madsen) è Sidewinder.
